荔枝果肉多糖之分离纯化与结构表征、抗氧化活性研究

第一章 绪 论

引言

荔枝(Litchi chinensis Sonn)属无患子科,古称荔枝、离支、丽支,果实成熟时果皮红艳诱人,故又俗称丹荔;果肉(实际上是荔枝的假种皮)为白色、半透明、香甜多汁、柔软而脆[3]。

荔枝为广东四大名果之一,俗有“岭南果王”之称,具有较高的营养价值和多种滋补养颜效果,一直被视为“果中珍品”。

我国是世界上栽培荔枝最早的国家,已知有 100 多个品种。

目前,亚热带地区气候和土壤合适的地方都有荔枝种植[4],我国荔枝栽培主要在广东、海南、广西、四川、福建、云南、台湾等省区。

在国外,越南、老挝、斯里兰卡、印度尼西亚、波多黎各、菲律宾、毛里求斯、马尔加什、南非、古巴等也产荔枝,但都是直接或间接从我国引入栽培的[5-6]。

年广东荔枝种植面积 400 万亩,产量达到 96 万吨, 年种植面积 413 万亩,产量达到 91 万吨,产量占全国总产量的 60%,面积为全国种植面积的一半[7]。

但是荔枝不耐贮藏,采后短时间内极易发生褐变和腐烂变质,加之产量的大幅增加与贮藏、运输和市场方面的制约,往往造成大量鲜果因未及时处理而败坏,屡屡出现丰产不丰收的现象。

所以,开展荔枝精深加工,开发荔枝功能性食品已迫在眉睫,这对于提高广东省农业生产效率,推动荔枝产业的持续发展具有重要意义。

本章阐述了荔枝及荔枝果肉多糖研究现状、国内外多糖研究进展并简要介绍了本课题的研究意义和研究内容。

荔枝及荔枝果肉多糖的研究进展

荔枝具有很高的营养和药用价值,据《开宝本草》记载,荔枝可止渴、美容。

荔枝全身都可入药,荔枝树根(行气止痛、固肾涩精)、树皮(疏肝解郁、祛风健胃)、树枝(治虚喘)、叶(解暑消滞、收湿敛疮)、

花(治喉痹肿痛)、果壳(治血崩)、果核(行气散结、祛寒止痛) 均可药用,荔枝果肉则是岭南佳果之一,适量食用,有补脾、益肝、养血、悦颜之功效[8]。

结合本课题我们主要讨论有关荔枝果肉营养成分和果肉多糖功能的研究概况。

荔枝果肉营养成分

荔枝果实中含有大量的糖分、维生素C、磷、钙以及少量的蛋白质、脂肪、维生素B等营养物质,荔枝果肉中矿物质元素含量也较丰富。

研究认为荔枝中的酸主要是苹果酸、琥珀酸和柠檬酸[9],而另有报道在荔枝果肉中采用相同的方法没有发现琥珀酸[10]。

荔枝果实的糖/酸比较高,存在品种差异,妃子笑为 ,糯米糍高达 [10]。

Chyau等[11]发现荔枝果肉的香气成分由游离态和结合态两种形态组成,游离态气味成分主要有三羟基-2-丁酮(),香叶醇()等;结合态气味成分主要为香叶醇()和香叶醛()。

刑其毅等[12]用冷冻收集法收集到的荔枝果肉头香水溶液研究荔枝香味成分,鉴定出 34 种香味成分,主要成分有醋酸、异戊醇、醋酸甲酯等。

荔枝果肉含有丰富的蛋白质和氨基酸[13];荔枝汁中氨基酸种类齐全(17 种)、含量高,每 100mL含 氨基酸,

除色氨酸外,含有人体所必需的 4 种氨基酸;总氨基酸中必需氨基酸的含量高达 44%,其中天冬氨酸、苏氨酸、丙氨酸、蛋氨酸含量较高[14]。

有关荔枝果肉的功能性有效成分的研究,国内外都有报道。

日本学者Minakata等报道荔枝果肉提取物在抗诱变方面具有较好的效果。

Shela等[15]研究发现荔枝鲜果中总的多酚含量为 鲜果,五倍子酸的含量为 ug/100g鲜果,这些成分可能对人体有保健功能。

第二章 不同荔枝品种多糖含量及不同干燥方法对其抗氧化活性的影响

引言

荔枝品种是由来自一个实生单株的营养繁殖系的群体组成。

通常认为生产种植广、栽培面积大、产量较高、适应性较强、品质较好的品种为优良品种。

荔枝品种在生产上具有一定的经济价值,且遗传组成基本相同,遗传性状稳定。

广东省作为我国荔枝的第一产区,种质资源之丰富为世界鲜见[1],目前已经收集和记载的优良品种、品系和单株就有 200 个以上,

其中作为商品性生产栽培面积 亩以上的主栽品种就有 26 个,包括:三月红、白糖罂、白蜡、状元红、园枝、大造、进奉、妃子笑、黑叶、怀枝、桂味、糯米糍、挂绿、雪怀子、红糯、香荔、尚枝等[2]。

目前还没有关于荔枝多糖含量与品种差异的报道,而且荔枝在品种上存在同名异物,同物异名及多名的现象比较严重。

所以,本章从广东省果树研究所荔枝资源库中选取经果树所专家鉴定的在广东省主栽的 21 个荔枝品种,研究其活性成分,以期弄清楚广东地区主栽荔枝品种间多糖含量的差别,为后续研究材料的选取提供依据。

荔枝生产季节比较集中,不容易保藏,所以大量荔枝需要通过加工消化。

目前,荔枝加工主要是荔枝干,占全部荔枝加工品的 80%以上[3]。

传统荔枝干的加工方法主要有日晒法和火焙法,这两种方法加工的荔枝干风味较淡,果肉颜色较深,烘干过程干燥不均匀等。

微波干燥起源于 20 世纪 40 年代,微波辅助干燥技术已经广泛应用于食品工业和农产品加工等领域[4]。

研究发现微波干燥与传统干燥方法相比较提高了干燥速率,减少了干燥时间,提高了工业产量和产品的品质[5-7]。

目前,微波干燥技术已被应用到农产品的干燥技术的研究中[8-12],在果蔬产品干燥方面,分别对荔枝[13]、龙眼[14-15]等进行了微波干燥研究。

材料与方法

荔枝原料

荔枝为采自广东省农业科学院果树研究所的新鲜荔枝。

所采摘的荔枝大小、形状和成熟度尽可能保持均一。

根据荔枝成熟时间及栽培面积,选取广东地区21个品种,其中早熟品种包括:称砣、白蜡、妃子笑、白糖罂、园枝、黑叶、大造、状元红8个品种;

中熟品种包括:丁香、桂味、鸡嘴、青皮、田岩、水晶球6个品种;晚熟品种包括:糯米糍、红糯、玉荷包、挂绿、怀枝、尚枝、雪怀子7个品种。

干燥方法试验采用荔枝品种为黑叶,采自广东省农业科学院果树研究所的新鲜荔枝。

所采摘的荔枝大小、形状和成熟度尽可能保持均一。

摘要:语用模糊是语言运用过程中产生的一种不可避免的现象。本文简明扼要地阐述了语用模糊的概念,产生,类型,功能和存在的意义,目的在于给读者提供一个语用模糊的基本概念并希望以此引起更多学者关注这一现象。

1. 语用模糊的概念

语用模糊指的是说话人在特定语境或上下文中使用不确定的,模糊的或间接的话语向听话人同时表达数种言外行为或言外之力(illocutionary acts or forces)的现象(Thomas, 1995; 俞东明,1993)。例如:

例(1)Would you like to come in and sit down?

这句话可以同时实施邀请,请求或命令等不同的言外行为,也可以是三种言外行为兼而有之,其界限是模糊的。

语用模糊比较常见的英文表述是“pragmatic ambivalence”(根据《英汉双解剑桥国际英语词典》“ambivalence”意指“矛盾情绪,矛盾心理”,笔者认为这里可能指语言结构形式进入运用中所遇到的各种矛盾),但是也有人称语用模糊为语用歧义,用“pragmatic ambiguity/ambivalence”来表达(项成东,;周红,)。笔者认为无论是“语用模糊”还是“语用歧义”,只要是从语用学角度,在语用学的范围内进行的研究,就都可以接受。

2. 语用模糊的产生

根据Thomas(1995:22) 的动态语用学理论,语义的产生与理解是一个动态过程,必须有以下要素的共同参与才能完成:说话人与听话人对意义的商榷讨论,话语产生的语境(包括物质,社会和语言等诸方面),话语可能表示的各种潜在意义。在此基础上,我们可以把语用模糊产生的原因分为以下几种(周红,2002):

指示语引起的语用模糊。指示语是指那些表示指示信息的词语,包括人称代词,指示代词,定冠词以及一些表示地点和时间的副词。指示信息不清楚的话语往往很难理解甚至会产生语用模糊。例如:

例(2)He said he was a teacher.

例(3)He said he was coming today.

会话涵义引起的语用模糊。在我们所说的话和我们说话的用意之间常有一定的距离,这种话语的用意就是“会话含义”(conversational implicature)(何兆熊,2001:152)。说话人的交际意图一般是隐而不宣的,不能靠所说话语的字面意义来确定,而要靠听话人根据语境进行辨认和推导,真正认识到说话人在说这句话时赋予话语的施事行为用意。例如:

例(4)Is that your coat on the floor?(可能是“指责”“命令”)

例(5)Waiter, there is a fly in my soup.(可能是“抱怨”“指令”)

间接言语行为引起的语用模糊。

Searle认为间接语言现象实际上是“通过实施另一种言语行为来间接的实施某一种言语行为”(1975:60),是一种以言行事(illocutionary act)。要理解间接言语行为,首先要理解“字面用意”(literal force),然后从“字面用意”推断出间接用意,即说话人所要表达的施事行为用意,这也是引起语用歧义的具体表现。例如:

例(6)Can you pass the salt?(可能是“询问”“请求”)

例(7)There is a wolf in the mountain.(可能是“陈述”“警告,吓唬”)
